what are the signs that show that the timing belt would need to be replaced?

I have a 2005 Lexus RX330 which I had the timing belt replaced at about 100,000 Km.
Now, there's a squeaking noise coming from the belts when the weather is cold when starting the car.
Currently there's 250km on the car. Would these squeaking noise be a sign that the timing belt needs to be replaced? What is the proper interval at which the timing belt should be replaced?

Probably the squeaking noise is coming from either the pulleys or the serpentine belt. That's the good news. The bad news is the timing belt should be changed every 150,000KM or 90,000 miles. So you're due for another change of that belt.

^^ +5 t-belt is due at 150,###kM/90,###mi...
Typically cogged belts do not squeak. Its the V and serpentine belts that squeak from a tiny bit of slippage on the pullies.
Temperature change induces condensation which when on the pulleys and belt allow it to slip and squeak. Then it goes away as the moisture is displaced.

To add to this my pulleys/idlers started to scream (literally) and water pump seeped coolant which was visible at the bottom of the timing cover. My belt was overdue due to age not mileage as I purchased the car which was 2005 with only 71K miles. Belt was in rough shape but to be honest I could have squeezed at least another 10K miles out of it no problem.
