Remove 140 Amp Alternator Fuse

Hello Everyone,
This is my wife's 2008 RX 350 with 203,000 miles
I changed the alternator (what a challenge) after it started smoking and the car died on the side of the road. As much as I'm ashamed to admit it, I bought a rebuild from AutoZone and had them bench test it before I left the store. I installed it and the battery light came on and the battery is not charging.
I checked the little 7.5 amp fuse and it is good.
I'd like to check the big 140 amp alternator fuse and I'm hoping someone can tell me how to remove the fuse? It's way down in the box and I can't seem to pull it out.
Does the fuse pull straight up and out?

P.S. The thought of having to pull the alternator again is making me a little ill.

The fuse is bolted in never worked on your car but the fuse block needs to be exposed (lifted out of the frame, or maybe not) so you can remove two bolts. The fuse looks like this
