Oh boy, sounds like you are in deep with this repair already.
What caused the overheat?
How hot did it get?
Did you diagnose the head gasket leak or just pulled the engine down looking for damage?
Compression test done?
Leak down test?
Got fuel at the rail?
Does it sound like it's trying to start?
Any codes?
Any more details that you think would help us figure out what's going on can help cuz it's impossible to guess without more info.

Just one?
Are you sure you have a Lexus RX 330?

Head gaskets on this V6 would be a major repair.
