I can't seem to get the doors to Auto Lock on my '12 RX350.
I went to settings and changed the time from 60 seconds to 30 seconds, they still Don't Auto Lock and my spouse keeps forgetting to Lock the doors when she gets out!
It's got a Keyless ignition.
Any Simple things I could try before I take it to the dealer?

My '05 RX330 has the Auto Door Lock feature, and relocks the doors after about a minute of inactivity.
I read here, that the 2012 RX doesn't have this feature?

If the car is off and locked and you unlock it from the outside but do not open the doors - it will relock.
I am not aware of any other situation where the car will auto lock for you while the engine is off.

There are little caveats to what RadiantX45 stated, but as a statement it is correct. Caveat being the keyFob should not be detected inside the vehicle.
So if you unlock with keyFob and do not operate the door handle it will lock itself again.
The caveat stated above, prevents vehicle locking out with the key inside.
If any door lock fails to respond, there is fail safe mechanism to unlock all doors. [This is my understanding as safety is allowing the owner to get inside the vehicle and manually lock doors vs left out in the cold]
Please check the manual as there may be some settings to enable auto lock doors. Mine is set on shifter going to "R" and unlock on "P". There may be options for doors other than the driver's.
Nit: You mentioned front 2 doors. The lock-switch and the keyFob should operate all doors.
