I have a new noise on our 2010 RX450h. When parking after shifting into park there is a loud squealing noise when releasing the brake.
Any ideas?

These noises are very difficult to diagnose over the internet. If you attached a recording that would help a lot with determining the possible source.
I suspect it is likely related to a stuck or frozen brake caliper causing rubbing. The caliper slide pins need to be greased every few years or they can become stuck. Since the hybrid brake pads can last 100,000 miles, these pins are often neglected. Lubing them is what is called servicing the brakes.
If we give you some ideas, will you try to fix it yourself or take it to a shop? If a shop, I recommend you find a well regarded independent shop as the dealer will just overcharge, especially for a diagnosis. A good independent shop could probably test drive your car and give a possible source of the noise in 5 minutes.

I edited your initial post to 2010. You may also want to check the hybrid forum here in case it is some weird hybrid only noise.

I have tried to rephrase [as best as I can understand] ... please correct as needed
1. When you stop in D with your foot on the brake there is no noise.
2. When you shift after stopping from D to P there is no noise.
3. When you lift the foot of the brake there is noise.
Pop the hood after 1 and have an assistant try to figure out or estimate where the sound is coming from ... a) engine compartment or at the wheels
Brake fluid needs to be replaced at certain time. Please check the maintenance requirements. Has that been done?
