The black caps are marked H for high and L for low. I think they may have different sized threads to reduce the chance of a mistake. I have not charged A/C in a long time so maybe someone else can assist. Or try YouTube.
imho ... you will be spending about $30 as a DIY attempt. The same service with diagnosis [assuming there is no additional issue] will cost you about $120 at an auto AC shop. The additional $90 is well worth the risk of doing damage in three figures. This is what I would do, but it is your vehicle and your money/saving.
I need to recharge my AC and I want to make sure that this is the low pressure port. Can someone please confirm?

The low pressure port and high pressure port are different sizes so the A/C recharge kit fitting will only fit the low pressure port. I never needed to refill my RX350 A/C so I can not verify if the port in the picture without the cap is the low pressure port, but the A/C recharge kit fitting will only fit on the one port which seems to be the one without the cap.
Note: Replace that cap to prevent the refrigerant from leaking out in the future. The cap has an o-ring to prevent the refrigerant that gets past the shrader valves from leaking out of the A/C system.
