Informal Poll on Tire models contributing to "stiffness"
I suggest those concerned with the stiffness of the 2010 RX ride on choppy roads add tire model info. I believe there are 3 tire models (18 or 19 inch) being distributed on 2010 models.

summer or "performance" tires definitely contribute the the stiffness. Increased cornering for sure, but at the cost of stiffer sidewalls.
My RX is AWD, has dunlop tires, 19". My opinion is they are quiet, corner well, but do ride stiffer on choppy roads than my previous 2004 RX AWD with Michelin MXv4 and subsequent replacement Bridgestone alenzas.

Springs do lossen up after a while, but probably not to the extend that you are looking for. Lexus made a trade off in the 2010 RX, they made the handling a bit better, but at the same time, made the ride stiffer.

executive matchup
i would recommend downsizing from your current 19" wheel package to say 17" wheel with a higher profile tire. this should solve your bumpy problems. and get a touring tire, they are supposedly more compliant on bumpy roads.

I have about 2400 miles. I have not sat in the rear yet, but going over the same bumps 5 days a week, I feel a huge improvement in reduced bounce. I agree with G-Man, give suspension some time to loosen up. I have 18" Bridgestone HL/400s, passenger tire.

I went to the dealer today and they couldn't find anything wrong with my car. They stated that it was performing normally. I took another 2010 RX 350 on a test drive, this one with 18" tires to see if I could feel any difference. I couldn't. There was no noticeable difference between my car and the one I test drove, even with the 18" tires. The only differences I felt were my car seems a little heavier (might be true or my imagination) and mine seems to handle slightly better (likely attributed to the 19" tires).
So if anyone is worried about getting the 19" tires because it will hurt the ride quality then I can safely say that it won't. However the sad thing is the "problem" we're facing isn't really a problem in the eyes of Lexus. It's designed to be that way. Likely as others have stated they made a trade off on the ride quality for better handling.
I'm hopeful that I'll see an improvement over time as others have stated. I have just over 1000 miles on mine, so we'll see how it is at 2000. I suspect there may be a break-in period but I doubt it will be a significant difference. Only time will tell! :-)

to bad they did not go back and put 16" on the 2010 then you have a lot more tire side wall and not all this junk going on about haveing it ride smoothly and the ride quality

that is still an option for you if you want to. nobody said you have to stay with 18" or 19" oem wheels
