Wireless key won't open back door
Is there a way to reprogram the wireless key? All functions work properly except for opening and closing the back door. Both keys. 2012 RX 350

OK, so we've established that the motor itself works fine and the activation process with the button from the cabin works fine, but not via either of the two remote keys.
Did it use to work and doesn't now?
If the vehicle is new to you, you might not know that you need to hold down that tailgate button on the remote until the hazard lights flash and the tailgate opens. Just pressing the button momentarily will not activate the motorised tailgate.

Back door, not tailgate, right? As it somehow steered towards tailgate, not rear door.
Check simple. Is child lock engaged? Though it would have prevented it from being unlocked from handle or button inside but hey...
Otherwise, as key does not differentiate between specific door, it opens all or none (my Dodge RAM open driver door with the first click, then all with the 2nd) - you have continuity problem from remote lock module to that particular door. Likely easiest is to remove door trim, get to the lock plug and see if 12V is triggered when remote button is pressed.

Have pressed and held the button on both keys. No dice. Car is new to me and the tailgate/trunk door has never opened with the key. Will check the other things suggested. Thanks.

I'm still confused, are you talking about the back doors or the rear hatchback ? If it's the hatchback door there is a button in the glove box that disables or enables that function, but even then you have to hold the key fob hatch button until the lights flick

There are few more details missing:
1. Release/lock. Do you hear that operating. Move the key-fob away and see if you can lock/unlock with the inside button. Then check if it locks/unlocks based on how you have programmed the RX (lock when you move out of Park).
2. Lift/close motor. Does that operate with the driver button. .. there is safety button in the glove compartment to disable/enable power lift/close
3. Now check the key-fob. Open- also unlocks the vehicle (may or may not be a programmable option).

Maybe the batteries in both the keyfobs are low and cannot sustain a constant transmit when that button is needed to be held down for a few seconds. My suggestion would be to replace at least one of the batteries with a fresh CR1632 battery. This will only cost a couple of dollars.

So now you need to clarify two more questions:
1. When you press and hold liftgate button on fob, does it open?
2. when you press Open button on fob, do all DOORS open but liftgate does not? As in - STAYS LOCKED?

Power back door

After all the doors have been unlocked, the rear door can be programmed for the following:
a. After momentarily pressing the rear door release button on the remote key, the rear door will open.
b. After pressing the rear door release button twice on the remote key, the rear door will open.
c. After pressing and holding the rear door release button on the remote key for 0.8 seconds, the rear door will open. (Standard)
d. The rear door, release door button is turned off and will not function
It is likely that your key has been programmed to either setting d, or maybe possibly setting b. You can test this out simply by double pressing the tailgate release button on your remote key.
