I guess you can update us as you are getting the water pump changed and the mechanic will hear this noise and investigate.
Could be a water pump bearing worn out, but I would expect you to be loosing coolant.
Best to remove the belt and spin each item by hand and feel for smoothness.
Idlers can wear out, check alternator, AC compressor.
Any chance you hit the bottom of the motor on a curb? might have hit a pulley.
Please update, hopefully it is just a waterpump. You did not say how many miles you have, you may as well put a new belt, you should just have to pay the cost of the belt, cause he would install your old belt anyways.

+1, wp, common. Search "water pump" on gen 2 RX350 forum (more high miles cars there), same engine.

That is the sound of a water pump with bad bearings. They can fail at any time. I had the exact same noise on my 2016 Toyota Sienna (same engine) with only 37,000 miles last Summer. It did not leak coolant and the noise is caused the internal seals on the water pump failing and allowing some coolant to remove the bearing grease and causing bearing failure. I did not want to deal with it due to the confined space and a trusted independent shop replaced it with a NAPA water pump (with a lifetime warranty) for $1100. They also installed a new serpentine belt.
