I thought it was fairly easy to do myself. As FlopTop04 mentioned, old card should be removed with vehicle off. You have to get the old card out by pushing on it, which makes it pop out. Getting the old card out was hardest part for me because it didn't pop out very far and my fat fingers couldn't grasp it to pull it out. Had to use a pair of tweezers to grab it but it came right out. Make sure the new card is pushed in all the way, then start the vehicle and it will load the new maps. You must leave the vehicle running while it does the update. Once that was done, a message came up instructing me to turn off the vehicle and then start it back up. After I did that, the process was finished and everything worked fine. The whole process took about 15 minutes.

You can go to any Lexus dealer to get one.
On my first Lexus with Navigation. I bought my first update from the dealer. It was the old CD Disc style.
Then I read that people who buy the update. Sell their last years updates on eBay. So, for the rest of the time I owned the car. I just bought last year's update for just a few dollars.
Not sure if that would work today with the small Computer card today.

That process worked on my SC430 with GEN 4 navigation but...
Pretty sure that trying to use a previously used MicroSD Map cartridge would not work with GEN 9. During the initialization / load process, the vehicle actually writes something on the cartridge that makes a permanent link between that car and that cartridge, rendering it useless on any other vehicle. Call it ensuring Copyright License.

It should be similar to this setup on GS and ES:
