Technically, yes.
The "power" button simply changes the shift point so that upshift points are selected at higher engine speeds. So I guess you would be working the engine a little harder. On the other hand, these engines are still going strong in 1990-91 LS400's with 200,000 miles and more on them.
I wouldn't worry about it. The bigger impact is probably on mpg, which, of course, is short term, and that wasn't your question.

Gas Mileage
Everyone's mileage is showing significant differences....why don't we survey this?...I think the other three factors we need to know are 1) Odometer Reading 2) Average MPH 3) State of residence and 4) ECT power switch position. This way we can compare apples to apples.
I am at 1800 total miles, getting from 14 to 16 MPG, with an average MPH of around 26MPH, my ECT is always on, and I am in Southern Califonria

I am at 13,000 miles, the skies are clear and bright, I generally have one passenger at about 100 lbs, who usually has a sunny disposition, the top is up about half the time, except at highway speeds, when it is always up, I use premium gas most of the time, seldom really step on it unless there is a hot chick on board -- I eat right except on the weekends when I tend to load up on carbs, which is probably not good for me. Also, when I drive my other car, the SC gets pissed and the engine tends to lag for a while, so that may have an impact. I am not in California.

I have 7k miles and consistently avg 21 mpg according to my computer, driving fairly aggressively always in Power mode, 50/50 hwy/city around Baltimore. I'm very satisfied with the mpg, especially considering my previous Audi TT 6-spd and BMW 528 didn't avg all that much better.

I am at approximately 19,000 miles (2002 SC, delivered May 2001). I drive mostly city, some highway. Average MPH is 16. Live in Southern California. Try to have top down as much as possible. Run the A/C when temps are over 80. I use premium fuel exclusively. I have had the oil changed regularly, and paid the Lexus dealer too much to perform the 15K service.
I'm averaging 14mpg. If I take a road trip on the freeway, I get 22mpg.
ECT switch is not switched to Power or Snow (it's in the neutral position).

13,000 miles. Mix of stop-and-go and open highway driving. Air conditioning always on. ECT mode with fairly conservative driving but occasional 2Fast2Furious outbursts. Consistent 18-19 mpg.

BTW: I'm at a measly 8K miles... but soon It'll jump about 6K for the out west road trip it's going on in early September...
Also, I live in the flatlands of the midwest... I think we are at 860 feet above sea level.... LOL
It will be interesting to see my mileage variance on the road trip in the rockies, desert, dry arizona climate, etc.

according to my computer on the car, I get 20-23 miles per gallon consistently on premium fuel. I'm at 6,100 ft altitude. Went out to Kansas for a trip. Man, it's amazing what oxygen can do for performance!! Time for a supercharger!!

18-20 mpg. Moderate driving, ECT always on, 75%city driving (3d & 4th gear), 25 % HWY (D) - top down as much as possible.
I am approximately 10 feet over sea level. With our typical summer humidity, I don't think the air can get much thicker!!!
